Emulsion Breaker
This emulsion breaker is a clear, fluorinated chemical solution specifically formulated to efficiently disrupt microfluidic droplets.
Fluo-Break enables easy recovery of the droplet phase and is fully compatible with Emulseo’s surfactants and oils (Surfactants for droplet stabilisation and Fluorinated oils). Fluo-Break is suitable for a wide range of applications, including cell recovery from microfluidic droplets.
Applications
- Diagnostics: digital droplet PCR (ddPCR) for DNA analysis.
- Biotechnology: Single-cell analysis for stem cell research and immune analysis.
- Pharmaceutical development: Screening of potential drugs.
- Encapsulation: Stabilisation of dyes or small molecules in droplets.
Protocol
- Transfer the emulsion into a microcentrifuge tube. The upper phase corresponds to the emulsion, while the lower phase contains the excess FluoSurf™ diluted in Fluo-Oil.
- Carefully remove the excess oil (bottom phase) with a pipette tip.
- Add Fluo-Break to the emulsion (1x emulsion volume) and close the lid.
- . Shake the microcentrifuge tube manually. The emulsion will begin to break.
- Centrifuge the tube for 1 minute. This will result in complete phase separation: the upper phase will contain the aqueous solution, while the lower phase will consist of a mixture of Fluo-Break and Fluo-Oil.
- Carefully collect the upper aqueous phase using a pipette and transfer it into a clean microcentrifuge tube. The recovered aqueous phase is now ready for downstream experiments.
